dc.description.abstract | Působení {α}1, {α}2, {β}3 adrenergních agonistů na klidovou spotřebu a působení {α}, {β}2 adrenergních antagonistů na noradrenalinem stimulovanou spotřebu kyslíku na izolovaných lidských leukocytech (PBMC) bylo studováno za použi Clarkové kyslíkové sondy. Bylo objeveno, že {α}1 agonista mírně zvyšil klidovou spotřebu kyslíku PBMC, zatímco aplikace {β}2 a {α} adrenergních antagonistů výrazně inhiboval noradrenalinem (NA) stimulovanou spotřebu kyslíku. {α}1 a {β}3 agonisté neměli žádný účinek na klidovou spotřebu kyslíku PBMC. Noradrenalinová termogeneze v PBMC je zprostředkována {β}1 adrenoreceptory (z 25%), {β}2 receptory (z 8%), a {α}2 adrenoreceptory (z 12 %). Noradrenalinová termogeneze PBMC je mediována jinými adrenergními receptory než NA termogeneze v adipocytech hnedé tukové tkáni.
NA termogeneze PBMC je nezávislá na přítomnosti organických substrátů v kultivačním médiu. Podání substrátů (glukosy, glutaminu, methioninu) do média ochuzeném o tyto substráty nemělo vliv na klidovou a NA stimulovanou spotřebu kyslíku PBMC. Přídavek cysteinu však zvyšoval NA stimulovanou spotřebu kyslíku PBMC a naznačuje tak, že acetyl-CoA může být jedním z důležitých substrátů pro NA termogenezi PBMC. | cze |

dc.description.abstract-translated | Effects of {α}1, {α}2, {β}3 adrenergic agonists on resting oxygen consumption and effects of {α} and {β}2 adrenergic antagonists on noradrenaline stimulated oxygen consumption of the human peripheral blood mononuclear cells (PBMC) were studied using the Clark oxygen electrode. It was found that, the {α}2 agonist increases slightly the resting oxygen consumption in the PBMC while, the {β}2 and the {α} antagonists inhibit the noradrenaline stimulated oxygen consumption considerably. The {α}1 and {β}3 agonists have no effect on the resting oxygen consumption. The NA termogenesis in the PBMC is mediated by {β}1 (25%), {β}2 (8%) and {α}2 (12%) adrenergic receptors and not by {β}3 adrenoreceptors. The NA thermogenesis in the PBMC is mediated by different adrenoreceptors than the NA thermogenesis in the brown adipose tissue.
The NA thermogenesis of PBMC is independent on the presence of organic substrates in the cultivation medium. Administration of external substrates (glucose, glutamine, methionine) to the medium has no effect on resting and NA stimulated oxygen consumption of PBMC, while administration of cysteine increases NA stimulated oxygen consumption considerably, indicating that acetyl-CoA could be an important substrate for NA thermogenesis in PBMC. | eng |
