dc.description.abstract | Extracelulární adenosin je významnou signální molekulou zapojenou do mnoha fyziologických a patofyziologických procesů. Efekt adenosinu je pravděpodobně zmírněn adenosinovými deaminázami a jeho transportem do intracelulárního prostoru pomocí nukleosidových transportérů. Mnohé studie na buňkách obratlovců i bezobratlých in vitro popisují inhibiční vliv adenosinu na proliferaci buněk. Mechanismus účinků adenosinu je však složitý a do značné míry specifický pro daný typ buněk a je zprostředkován buď aktivací specifických adenosinových receptorů, nebo transportem adenosinu do buněk. Tato práce sledovala mechanismy toxicity adenosinu u buněčných linií Drosophila melanogaster. Experimenty ukázaly, že adenosinová toxicita korelovala s mírou transportu adenosinu do buněk, jeho fosforylací adenosinovou kinázou a inkorporací do ATP. Metabolické účinky adenosinu včetně zvýšení glukózy v buňkách byl nezávislý na aktivaci AMP kinázy. | cze |

dc.description.abstract-translated | Extracellular adenosine is a multi-potent signalling molecule implicated in many physiological and pathophysiological conditions. The level of adenosine signalling seems to be attenuated by te action of adenosine deaminases and adenosine uptake into the intracellular space via nucleoside transporters. From many in vitro studies on both vertebrate and invertebrate cell lines, adenosine is known to reduce cell survival. The mechanism of adenosine action seems to be complex and largely cell type specific resulting from either activation of specific adenosine receptors or adenosine uptake into cells. This study aimed to elucidate the mechanisms of adenosine toxicity in fruit fly cell lines. Experiments showed that adenosine toxicity correlated with the level of adenosine uptake, its adenosine kinase mediated phosphorylation and incorporation into cellular ATP pool. Metabolical effects of adenosine included elevation of cellular glucose level and were independent of AMP kinase activation. | eng |
