The aim of the thesis was to gain a better understanding of decorin binding proteins A and B (DbpAB) of various Borrelia species my different means. Overexpression of DbpA or DbpB in B. afzelii was addressed, examining phenotypic changes in mutants. Also, it was aimed to create shuttle vectors to complement DbpAB of different European Borrelia species as well as suicide vectors.cze

dc.description.defence
the defense was held in a hybrid format - online and in person. Ms. Danklmaier was present online. Prof. Grubhoffer welcome the student, members of the commission, and the audience.

Ms. Danklmaier presented the basic information about the studied organism, Borrelia burgdorferi, tissue tropism, and surface proteins of the various Borrelia strains. Decorin binding proteins A and B were described in more detail. The aims of the thesis were presented followed by the results o the expression of the studied proteins, growth of bacteria, production of chimeric bacteria, knockout of Dbp proteins in Borrelia.

Supervisor Dr. Rego presented his review, followed by the opponents Dr. Langhansová and Dr. Lieskovská. Ms. Danklmaier answered the questions. Discussion and questions from the members of the commission and the audience followed. Ms. Danklmaier answered questions on the success and controls in the transformation of Borrelia and the cloning systems.

Voting on the grade was done using MS Forms.

6x excellent, 1x very good = the final grade is
cze
