dc.description.abstract | Fragmentace a zmenšování populací ztrátou habitatů je typickým znakem probíhajícího 6. masové vymírání druhů, z hlediska ochranářské genetiky je nutné porozumět změnám, které s sebou přináší. Pro budoucí rozhonutí v ochranářské genetice je podstatné porozumět vlivu velikosti populace na variabilitu imunitních genů. Abychom lépe porozuměli genetické variabilitě nestačí analyzovat pouze sekvence DNA, ale je potřeba zaměřit se na funkční proteinovou variabilitu. Tato práce zkoumá fyziochemické vlastnosti a struktury jednotlivých haplotypů Toll-like receptorů v alopatrických populacích galapážských drozdců (Mimus sp.). Toll-like receptory hrají klíčovou roli v imunitním systému, kde tvoří můstek mezi vrozenou a adaptivní imunitní odpovědí, detekcí patogenů. Data v této práci odhalují variabilitu jejich diversity napříč malými galapážskými populacemi a velkou kontinentální populací drozdce mnohohlasého (M. polyglottos). Porovnání heterozygotnosti populací přineslo zjištění, že míra zastoupení heterozygotů je menší v galapážských populacích než v kontinentální populaci. To naznačuje větší vliv diversifikující selekce v kontinentální populaci, nebo redukci genetické variability v galapážských populacích vlivem efektu zakladatele a menší populační velikosti. Kromě popisu genetické diversity byly popsány fyziochemické vlastnosti objevených haplotypů, vymodelovány a porovnány struktury všech haplotypů TLR1B, TLR4 a TLR15. | cze |

It is essential to understand the impact of population size on the variability of the immune genes for the future in conservation genetics. In order to better understand genetic variability, it is not enough to analyze DNA sequences only, but focus on functional protein variability. This paper examines the physiochemical properties and structures of the individual haplotypes of Toll-like receptors in allopatric populations of Galapagos mockingbirds (Mimus sp.). Toll-like receptors play a key role in the immune system, where they form the link between innate and adaptive immune responses, by the detection of pathogens. The data in this work reveal the variability of their diversity across the small Galapagos populations and the large continental populations of the northern mockingbird (M. polyglottos). Comparison of heterozygosity of populations revealed that the proportion of heterozygotes is smaller in galapagos populations than in the continental population. This suggests a greater impact of diversifying selection in the continental population, or a reduction in genetic variability in Galapagos populations due to founder effect and smaller population size. In addition to the description of genetic diversity, the physiochemical properties of the discovered haplotypes were described, the structures of all TLR1B, TLR4 and TLR15 haplotypes were modeled and compared. | eng |
