| dc.description.abstract-translated | The codling moth, Cydia pomonella (Lepidoptera) is a major pest species of pome fruit, causing great economic losses in the temperate regions of the world. Investigation of the sex determination genes in pest species is important as it can provide new targets for insect pest control. This thesis focuses on the analysis of two sex determination genes, Masculinizer (Masc) and doublesex (dsx) in the codling moth. The relative expression of Masc was measured during early embryo development in females and males using quantitative reverse-transcription PCR (RT-qPCR) while simultaneously the sex-specific splicing of dsx was investigated by reverse-transcription PCR (RT-PCR). The results obtained by RT-qPCR showed higher Masc expression levels in females than in males, contrary to the pattern observed in other lepidopteran species. The splicing pattern of dsx shows that sex is determined during the window studied (12-20 hours past oviposition). Therefore, these results indicate a different mechanism of sex determination from the one identified in the silkworm, Bombyx mori. | eng |
